Poplar complete leaf tissue analysis

Abstract

The leaf samples are collected in the month of July from poplars grown at GLBRC main site. The samples are analyzed for different plant nutrients (carbon, nitrogen, aluminum, copper, iron, manganese, zinc, boron, sodium, calcium, magnesium, potassium, phosphorus, sulfur). The main aim of study is to determine nutrient efficiency of trees grown under two different practices; fertilized versus unfertilized.

Protocol

• Select five poplar trees each from main and microplot areas of the five replicates. From each tree, collect five recently matured leaves. Discard leaves with any visible soil or chemical residues. Avoid collecting leaves showing any disease or insect injury or nutrient deficiency symptoms.
• From each plot, make two bags; one bag for main (25 leaves) and one for microplot (25 leaves). In total,10 bags from 5 plots will be collected. Bring leaves to laboratory and wash them thoroughly with water.
• Dry leaves at 60 degrees C for 48 h and ground dried plant material through a 1mm screen using a Christy-Turner 8” Lab mill.
• Weigh ground leaf samples (3-5 g) and send samples to MSU Soil and Plant Nutrient Laboratory for complete leaf tissue analysis.
• Carbon and nitrogen are analyzed at Kellogg Biological Station.
